Protein production

We help you select the most suitable expression system for your protein. Our goal is to maximize protein yield and solubility, supporting the best possible downstream results. We support both small- and large-scale expression, with the option for proteins to be overexpressed in the cytoplasm or secreted. The resulting pellets or conditioned media can be processed by our team or handed over to you.

Bacterial expression (E. coli)

Escherichia coli is a popular choice for protein expression because of its fast growth rate, cost-effectiveness, and ease of genetic manipulation. It can produce large quantities of recombinant protein in a relatively short time, making it well suited to high-throughput applications. This system is particularly suitable for proteins that do not require complex post-translational modifications. We recommend starting with a small-scale expression test to assess expression levels, solubility, and overall protein quality. During this feasibility test, we can screen several E. coli strains and compare two expression methods: autoinduction and IPTG induction. Once the best conditions have been identified, we scale up expression according to your needs.

Baculovirus expression (BEVS)

The baculovirus expression vector system (BEVS), commonly referred to as insect cell expression, is a powerful platform for producing proteins that require complex folding and post-translational modifications. It is therefore often a better choice than bacterial expression for eukaryotic proteins. In a small-scale setup, we first aim to identify the optimal conditions for protein production. We screen the insect cell lines Sf9 and Hi5, as protein yield and quality can vary between them. We also perform a time-course experiment, harvesting on multiple days to determine the optimal expression period. For Hi5 cells, we additionally screen two expression temperatures. In addition to providing pellets and conditioned media, we also offer virus production services.

Mammalian expression (HEK 293)

This expression system closely mimics the cellular environment of mammalian cells, helping to ensure that proteins are processed correctly. As a result, it is well suited to the production of therapeutic proteins, glycoproteins, and other proteins that require mammalian-specific modifications. Before scaling up protein expression, we conduct a time-series experiment to determine the optimal harvesting time, assessed by Western blot analysis. For GFP- or mCherry-tagged proteins, fluorescence-based analysis is also available using CellDrop, microscopy, or flow cytometry. We work with human embryonic kidney cells derived from the 293 cell line (HEK 293). Our setup supports both transient expression and the generation of stable cell pools.

We also offer Maxi prep services, either as part of the expression workflow or as a standalone service. Our AmMagQuatro Maxi automation system produces high yields of transfection-ready plasmid DNA.
