Protein production

We assist you in selecting the most suitable expression system for your protein. Our goal is to maximise protein yield and solubility, ensuring optimal downstream results. We support both small- and large-scale expressions, with the option for proteins to be overexpressed in the cytoplasm or secreted. The resulting pellets or conditioned media can be processed by our team or handed over to you.

Bacterial expression
Escherichia coli is a popular choice for protein expression due to its fast growth rate, cost-effectiveness, and ease of genetic manipulation. It can produce large quantities of recombinant proteins in a relatively short amount of time, making it ideal for high-throughput applications. This expression system works especially well for proteins that do not require complex post-translational modifications.

We recommend starting with a small-scale expression test to assess expression levels, solubility, and overall protein quality. During this feasibility test, we can screen several E. coli strains and two expression methods: autoinduction and IPTG. Once we identify the best condition, we scale up the expression according to your needs.

Baculovirus expression
The baculovirus expression vector system (BEVS), commonly known as insect cells, is a powerful tool for producing proteins with complex folding and post-translational modifications, making it a better choice than bacterial expression for eukaryotic proteins.

In a small-scale setup, we first aim to identify the optimal conditions for protein production. We screen insect cell lines Sf9 and Hi5 since they often produce different results depending on the protein. Additionally, we perform a time-series experiment, harvesting at multiple days to determine the ideal expression duration. For Hi5 cells only, we screen two expression temperatures.

Besides pellets and conditioned media, we also offer virus production services. 

Mammalian expression
This expression system closely mimics the cellular environment of mammalian cells, ensuring that proteins are properly processed. As a result, it is ideal for producing therapeutic proteins, glycoproteins, and other proteins requiring mammalian-specific modifications.

Before scaling up protein expression, we conduct a time-series experiment to determine the optimal harvesting time, which is evaluated by Western blot analysis.

We work with human embryonic kidney cells derived from the 293 cell line (HEK293). Our setup allows for both transient expression and generation of stable cell pools. Under request, isolated Maxi prep services are also available.
