Example requests
The following examples illustrate how a typical request might be scoped. These are starting points; we are happy to adapt the scope to your specific needs.
Aggregation and oligomeric state check:
Goal: Determine the size, homogeneity, and oligomeric state of your protein in solution
Requirements: Purified protein sample (≥ 100 µg)
Result*: Molar mass, oligomeric state assignment, size distribution data, and an interpretation report
Methods: SEC-SLS and/or DLS, depending on the question and sample
Stability profiling and buffer screening:
Goal: Assess thermal and colloidal stability, and identify buffer conditions that improve protein behaviour
Requirements: Purified protein sample (≥ 50 µg, ≥ 0.5 mg/mL per condition); buffers or additives to screen (or we can suggest a panel)
Result*: Melting temperature (Tm), aggregation temperature (Tagg), unfolding curves, and a ranked summary of conditions tested
Methods: nDSF and/or DLS, as required
Secondary structure and folding assessment:
Goal: Obtain a secondary structure readout and detect condition- or ligand-dependent conformational changes
Requirements: Purified protein (≥ 50 µg, in a UV-transparent buffer)
Result*: CD spectra, and if requested, a thermal melt overlay
Methods: CD
Binding characterization:
Goal: Measure binding affinity and compare interactions across variants and conditions
Requirements: Purified samples of both binding partners; please contact us in advance to discuss sample requirements and experimental strategies
Result*: Binding curves, affinity values (KD), and an analysis report
Methods: ITC, MST (Monolith X), and/or GCI, based on sample properties and question
Protein quality control (PQC):
Goal: A comprehensive assessment of purity, homogeneity, and solubility
Requirements: Purified protein sample + completed PQC submission form
Result*: Purity estimates, size distribution, thermal stability data, and a consolidated QC report
Methods:
Level 1: UV-Vis, SDS-PAGE, SEC-SLS, intact MS,
Level 2: UV-Vis, SDS-PAGE, SEC-SLS, intact MS, DLS, nDSF
